Abstract

A method is described for the preparation of 35S-labeled p-hydrazinobenzenesulphonic acid from commercially available [35S]sulphanilic acid. This reagent can be used for preparing labeled nucleoside or oligonucleotide derivatives by reacting it with the dialdehydes produced by periodate oxidation. A simple method for using the reagent is described together with methods for separating and assaying the labeled products. These methods are applicable to subnanomole quantities of nucleosides but the sensitivity could be increased considerably. © 1976 Academic Press, Inc.
