Abstract

We report our recent findings on pH-sensitive, ortho ester-based cationic lipids and their lipoplexes for gene delivery. At higher pH, such lipids were relatively stable and readily condensed with plasmid DNA into cationic lipoplexes. At lowered pH, such lipids rapidly hydrolysed, which in turn triggered their lipoplexes to destabilize the endosome membrane.
