Abstract

3,3′-Iminodipropionitrile (IDPN) is a synthetic neurotoxicant. The purpose of this study was to observe the effects of IDPN on rat behavior and LDH activity in brain and spinal cord homogenates. One group of rats was given a single dose of saline, 50 mg/kg IDPN, or 300 mg/kg IDPN. An open field test, a landing foot spread test, and a righting reflex test were performed before and after dosing. Rats were sacrificed at 2 hours, 48 hours, or 9 days post exposure and LDH activity measured in brain and spinal cord tissue homogenates. In behavioral testing, landing footspread of male rats in the high dose group was significantly different from the controls in the second post-dosing test. In LDH activity assays, brain and spinal cord tissues from male rats showed a significant decrease in LDH activity 48 hours (in the high dose groups) and 9 days (in both the low and high dose groups) post-exposure. Brain tissue from female rats showed a significant increase in LDH activity in the high dose group after 9 days of exposure.
