Correlation of iNOS, eNOS, PPAR{gamma}, TNFα and HNF4α gene expressions in subcutaneous adipose tissue of obese women in response to 12-week calorie-restricted diet with or without exercise

Abstract
OBJECTIVE: Determine the effects of weight reduction by a 12-week calorie- restricted diet with or without exercise on iNOS, eNOS, PPAR{gamma}, TNFα and HNF4α gene expressions in subcutaneous adipose tissue of obese women. RESEARCH DESIGN AND METHODS: Twelve women volunteers with BMI >30 kg/m2 participated in 2 separate experimental trials. The trials differed only by whether the women performed diet (D) or diet plus exercise (DE). In the DE, aerobic exercise was performed 3 days per week (30'/session). Biopsy of subcutaneous adipose tissue before and after the program was processed by Real Time-PCR for all genes. RESULTS: The relative expression of mRNA by iNOS, eNOS and PPAR{gamma} genes showed a significant decrease in the D, p=0.028, p=0.046 y p=0.028 respectively. A decrease in the iNOS (p=0.028) and PPAR{gamma} (0.075) gene expressions was observed in the DE. TNFα and HNF4α genes was not significant differences. We found a positive correlation between the iNOS and eNOS gene expressions and the BMI (SC=0.771) for both groups. CONCLUSIONS: A caloric restricted diet (D) and (DE) have an influence on iNOS and eNOS gene expressions. The positive correlation between PPAR{gamma} and TNFα mRNA expression shows that these transcriptional factors may regulate the NOS expression in these women. These genes could be therapeutic targets for new drugs in obesity treatment.
