AbstractRecently we described a novel strategy to render pH-sensitive lipid amphiphiles and their colloids: a protonation-induced conformational change of the built-in trans-2-aminocyclohexanol moiety. In liposomes, this ring flip loosens packing of the attached lipid tails, leading to contents leakage. Herein we report our latest studies on the pH-sensitivity of various PEG-grafted (sterically hindered) liposomes comprising trans-2-aminocyclohexanol-based lipids. These liposomes are stable at pH 7.4 and yet release their content in a few seconds at pH 5.3-5.5.
