Biochemistry Project Lab in Protein Purification Incorporating Learning Teams

Abstract
A project laboratory was developed for first semester biochemistry students demonstrating protein isolation, purification, and analysis techniques. The protein of interest was the iron containing mitochondrial protein cytochrome c. Through learning teams, students were introduced to isolation, purification, and protein detection techniques including homogenization, centrifugation, ion-exchange and size-exclusion chromatography, SDS-PAGE, and protein assays. Methods for the laboratory procedures were adapted from various published works to conform to time constraints of the student three-hour lab period. Instructors used formative assessment and undergraduate research students to trouble-shoot and optimize student laboratory sessions. Modifications and improvements to the published procedures will be outlined. Laboratory procedures and learning team logistics will also be available.
